The DOG1 (M) [DOG1.1] antibody is a mouse monoclonal antibody raised for the detection of DOG1, a cell surface protein of unknown function that is selectively expressed in gastrointestinal stromal tumors (GIST). DOG1 expression has been reported to be a highly sensitive and specific marker for GIST in formalin-fixed, paraffin-embedded tissue.15 Studies have demonstrated that among GIST cases with KIT mutations, DOG1 detected 11% more cases than CD117.16 In cases of GIST that are KIT/CD117 negative or carry PDGFRA mutations, DOG1 has been shown to improve diagnostic accuracy.16 Due to its localization on the cell membrane, limited expression in normal tissue, and strong expression in GIST tissue, DOG1 has been proposed as an additional target in both the diagnosis and potential treatment of GIST. 15-18 DOG1 immunoreactivity is less commonly observed in mesenchymal and epithelial tumors, seminomas, and melanomas when compared with CD117, supporting its specificity for GIST.15-18
