Glucagon, a 29-amino acid peptide, is secreted from the α-cells in the pancreas and from the endocrine cells in the gastrointestinal tract.15 Glucagon-secreting alpha-cells and insulin-secreting beta-cells make up the majority of the endocrine cell population within the pancreatic islets, and they work together to maintain glucose homeostasis.16 Glucagon works by binding to its glucagon receptor (GCGR), which is expressed in various organs, including liver, kidney, brain, intestinal tract, pancreas, and adipose tissue.15,16 Studies have shown that Glucagon acts as an activator of cancer cell growth through deactivation of Adenosine Monophosphate-Activated Protein Kinase (AMPK) and activation of Mitogen-Activated Protein Kinase (MAPK) in a GCGR-dependent manner. High levels of Glucagon may indicate the presence of glucagonoma, a malignant pancreatic tumor.17
